I'm getting constant crashes since some time, if i double-click .blend files. (Not from within Blender itself)
Nearly half of my old Blends crashes this way. It also happens if i Drag n Drop .blend on running Blender - crash.
Happens with 2.65, 2.64, 2.62
only if opened within Blender they work.

I now can see it does NOT crash if i remove my startup.blend from ~/Library/Application Support/blender/2.64/config
(but i'm not intending of redoing startup.blend just again, link all nodegroups and so...)
i recall i've had a very similar bug before, but can't find it... (old search function in bug trackes was waaaaay better than this google sht)

Can not reproduce the issue on linux. Perhaps something platform-specific. Does the crash happen with testbuil1 from http://download.blender.org/release/Blender2.65/ ?

It definitely looks like an OS X specific issue from the backtrace. I wasn't able to redo it yet on OS X 10.7, set the startup.blend and tried opening about 25 different files with own build and 2.65 test build, no crashes.
Are there particular .blend files where it always happens? Or is it just randomly crashing on any .blend file?

But it only happens if .blend is double-clicked or drag n dropped to Blender.... And only if my startup.blend is in ~/Library/Application Support/blender/2.64/config

Right, double click is what I'm trying, with your startup.blend. I noticed there are some addons installed that I don't have, could you try moving away the scripts/addons folder temporarily and but keeping your startup.blend, and see if it still crashes? Then we know if anything in the addons is triggering this.
Further it would be good if you could upload such a .blend that crashes when opening, I tried many different files here but perhaps there is still something specific about it that I don't have here.

The Attachement AluGerust.blend is such a file that crashes.

Thanks for the .blend file, tried it here but unfortunately no crash in any version I tested. Maybe it is a problem specific to OS X 10.8, though seems not that likely.
I noticed the startup.blend starts fullscreen, but then it goes to windowed mode when loading the file. Maybe that's related somehow, not sure.

Tried some more stuff. Removing Folders from ~/Library/Application Support/blender/2.64/
it only happens if the /Config is in there, files there are:
bookmarks.txt
luxrender.cfg
mitsuba.cfg
recent-files.txt
startup.blend
appended my /Config Folder

Thanks for the files, but still no crash here. I'll upgrade to 10.8 and see if I can redo it then.

Finally after upgrade I was able to redo the problem, so it seems to be a problem specific to this OS X version.
